How can I automatically (or manually) let tMM remove entries from the movie / series list which are not existing anymore. When I move movies / series form a directory to another, e.g. from an internal drive to an external one, tMM still shows the entries of the moved movies / series from the internal drive.

well, until you don't press "update datasource" TMM will do nothing
